mRNA-seq, or mRNA-targeted RNA sequencing, is a next-generation sequencing (NGS) technique used to study the transcriptome - the set of mRNAs present in a given cell or tissue at a particular time. It provides a global view of gene expression, including quantitative and qualitative information about mRNA.

RNA-seq workflow

01RNA isolation:  First, total RNA is isolated from the biological sample. The sample is then selectively enriched for mRNA through polyA enrichment or ribosomal RNA (rRNA) depletion.
02cDNA library preparation:  mRNA is converted into complementary DNA (cDNA) using reverse transcriptase.
03Sequencing:  The cDNA is fragmented, indexed and sequenced on an Illumina platform.
04Bioinformatics analysis:  The sequencing results are analysed to identify genes and their expression levels, as well as novel transcripts, isoforms or mutations.

Applications of RNA-seq

01Molecular biology and genomics:
  • Identification of gene expression patterns under different biological conditions (e.g. comparison of healthy and diseased tissues or treated and untreated subjects).
  • Discovery of novel genes, alternative isoforms and regulatory genes.
02Medical research:
  • Oncology:  Analysis of tumour transcriptomes to identify diagnostic, prognostic or therapeutic biomarkers.
  • Genetic diseases:  Understanding the molecular mechanisms of inherited diseases.
03Pharmacology:
  • Identification of targets for new drugs.
  • Monitoring drug effects at the level of gene expression.
  • Study of the activation and function of immune cells, such as T and B lymphocytes.
04Biotechnology and agriculture:
  • Understanding growth and adaptation processes in plants and microorganisms.
  • Improvement of agronomic or industrial-microorganism traits.

Technical specifications of mRNA-seq (polyA)

  • Library preparation: mRNA molecules with poly(A) tails are isolated from total RNA using magnetic beads coated with oligo(dT), which bind the poly(A) tails at the ends of mRNA. Total RNA is mixed with the beads, and after poly(A)-mRNA binding, the remaining RNA types (rRNA, tRNA) are removed by washing.
  • Library preparation options:
    • Stranded - strand-specific library preparation. Strand orientation information is retained during sample preparation, making it possible to determine which DNA strand each RNA transcript originated from.
    • Non-stranded - library preparation without strand specificity. In this approach, RNA transcription direction is not retained, resulting in the loss of strand orientation information.
  • Sequencing mode: 2x150 bp
  • Sequencers: Illumina platform, NovaSeq 6000 and NovaSeq X sequencers.
  • Recommended data output per sample:
    • 6 Gb per sample
    • 10 Gb per sample
    • 15 Gb per sample
    • Other configurations are also available depending on the client's needs.
  • Data quality: Q30 ≥ 85%
  • Recommended Sample Requirements - DNA/RNA:
